Sedimentation velocity (SV) AUC separates size variants in a sample based on differences in their sedimentation coefficients, which are a function of both the variant’s molecular weight and shape. The ability to achieve separation in a wide variety of solution conditions, without a stationary phase, with enhanced resolution, has made SV-AUC an attractive orthogonal method to traditional size separation methodologies such as size exclusion chromatography (SEC), and polyacrylamide gel electrophoresis (PAGE).
SV-AUC is recognized as a powerful tool for determining size distributions of macromolecules and quantifying protein aggregation in liquid samples. SV-AUC can also be used to obtain information about the hydrodynamic shape, sedimentation coefficient, and molecular weight of macromolecules.
